ATLAb™ - Antibody Humanization

BioAtla scientific team has developed a novel approach to rapidly humanize antibodies called ATLAbâ„¢ Antibody Humanization which is different from and superior to other humanization methodologies. The technology entails synthesizing a combinatorial library comprising the six CDRs of a non-human target monoclonal antibody fused in frame to a pool of individual human frameworks that our scientists have assembled. Our human framework library contains genes representative of all known heavy and light chain human germline genes. The resulting combinatorial libraries are then screened for binding to antigens of interest. This approach allows for the selection of the most favorable combinations of fully human frameworks in terms of maintaining the binding activity to the parental antibody. This process significantly increases the success rate relative to other technologies. In addition, the execution of our technology is simple as it does not require any a priori antibody structural knowledge, CDR analysis or framework design; therefore, the overall humanization process is greatly streamlined and cost effective. Humanized antibodies may then be further optimized using ATLAbâ„¢ Evolution technologies.
